- 【Abstract】Objective: Double-stranded RNA (dsRNA) has been shown to interfere with the function of specific genes in various species. The current study was design to
investigate whether short interfering RNAs(siRNAs) ofβ-site APP cleaving enzyme (BACE) can inhibit the expression of BACE in mammalian cells. Methods: To clone the gene of EGFP, U6 promoter andβ-secretase targeting siRNA by PCR, respectively. The PCR products were inserted into the retrovirus plasmid pLXSN. The interfering vector named pLXSN/EGFP-U6-siBACE. To produce the SK-N-SH cell line which can highly expressed BACE. Fluoroscopy and immunohistochemistry method to investigate the effect of BACE siRNA inhibits the BACE expression. Furthermore, to examine the production of Aβ. Results: The innterfering vector named pLXSN/EGFP-U6-siBACE was constructed successfully. The BACE siRNA can inhibit the expression of BACE in the SK-N-SH cell specifically and effectively,and reduced the production of Aβ.Conclusion: BACE siRNA inhibits the expression of BACE gene of mammalian, it provides an important experimental base for the application of RNA interference technology to prevention of AD.
